A 12-week double-blind, placebo-controlled study examined NMN supplementation in healthy middle-aged adults. Participants taking NMN showed a trend toward improved arterial flexibility, measured using brachial–ankle pulse wave velocity (baPWV), a widely used indicator of blood vessel health. Higher baPWV has been linked to an increased risk of cardiovascular events, making this a clinically meaningful finding. Further evidence comes from a 10-week clinical study involving overweight, postmenopausal women with prediabetes. Participants who took 250 mg of NMN daily showed improvements in how their muscles responded to insulin compared with those taking a placebo. NMN has also been studied for its potential role in supporting healthy ageing and longevity. As we age, levels of NAD+ naturally decline throughout the body, a change linked to reduced cellular function and the development of age-related health concerns. Research suggests that NMN may help support DNA repair, healthy blood flow and vascular function, while also assisting in maintaining muscle strength, metabolic balance and healthy gene activity as the body ages. In a randomised, multicentre, double- blind, placebo-controlled clinical trial involving 80 healthy middle-aged adults, participants received NMN or a placebo daily for 60 days. Those taking NMN showed no increase in blood-based biological age over the study period, while an increase was observed in the placebo group. This difference was seen across all NMN dosage levels (daily doses of 300 mg, 600mg, 900mg; noting that the TGA-recommended maximum daily dose is 500mg), suggesting NMN may play a role in supporting biological stability as we age.
