On the Horizon: What is NMN?

Dr Brad McEwen explores NMN and why this emerging longevity nutraceutical is gaining attention for its role in healthy ageing and metabolic health

What it is NMN? Nicotinamide mononucleotide (NMN) is a naturally occurring compound in the body that has become a key focus in healthy ageing and metabolic health. NMN helps the body produce NAD+, a vital molecule needed for everyday cellular functions such as energy production, cell repair, and healthy communication between cells. As we age, NAD+ levels naturally decline, and this drop can be accelerated by conditions linked to heart and metabolic health. Lower NAD+ levels are associated with reduced energy efficiency, increased oxidative stress and inflammation. By supporting NAD+ levels, NMN is thought to help maintain healthy metabolism, energy production, blood vessel function, and inflammatory balance as we age. How it works: Early research suggests that NMN may help reduce inflammation and oxidative stress in the body while supporting healthy energy production at a cellular level. It may also assist with blood sugar balance and insulin function, while helping to support healthy blood

vessels, circulation and heart health, including maintaining flexibility in the arteries. Additional findings point to potential benefits for brain health, particularly in areas such as memory and learning. The anti-ageing game-changer

levels of NAD+ in the body, alongside reductions in body weight, diastolic blood pressure, and total and LDL (‘bad’) cholesterol, compared with placebo. A 12-week double-blind, placebo-controlled study examined NMN supplementation in healthy middle-aged adults. Participants taking NMN showed a trend toward improved arterial flexibility, measured using brachial–ankle pulse wave velocity (baPWV), a widely used indicator of blood vessel health. Higher baPWV has been linked to an increased risk of cardiovascular events, making this a clinically meaningful finding. Further evidence comes from a 10-week clinical study involving overweight, postmenopausal women with prediabetes. Participants who took 250 mg of NMN daily showed improvements in how their muscles responded to insulin compared with those taking a placebo. These findings suggest NMN may play a supportive role in helping the body manage blood sugar more effectively, particularly in individuals with early metabolic changes. NMN is generally well-tolerated and considered safe in short-term human trials. As we age, levels of NAD+ naturally decline throughout the body, a change linked to reduced cellular function and the development of age-related health concerns. Research suggests that NMN may help support DNA repair, healthy blood flow and vascular function, while also assisting in maintaining muscle strength, metabolic balance and healthy gene activity as the body ages. In a randomised, multicentre, double- blind, placebo-controlled clinical trial involving 80 healthy middle-aged adults, participants received NMN or a placebo daily for 60 days. Those taking NMN showed no increase in blood-based biological age over the study period, while an increase was observed in the placebo group. This difference was seen across all NMN dosage levels (daily doses of 300 mg, 600mg, 900mg; noting that the TGA-recommended maximum daily dose is 500mg), suggesting NMN may play a role in supporting biological stability as we age.

• Perimenopause and menopause: During the transition to menopause – defined as the slowing and eventual cessation of production of the sex hormones oestrogen and progesterone - many women experience distressing symptoms, ranging from hot flushes and mood swings to vaginal dryness and breast tenderness.

It may be due to fibroids or polyps, or a side effect of using an intra-uterine device. An underactive thyroid may also trigger heavy periods, as can the approach of menopause.
