Results:
Baseline characteristics were balanced between arms. The primary objective was to compare the incidence of RTOG grade ≥ 3 mucositis. In the intention-to-treat analysis of 119 patients, grade ≥ 3 mucositis occurred in 11.7% of the celecoxib arm versus 16.9% of the glutamine arm, with no significant difference ( χ² = 0.678; p = 0.41; RR 0.69, 95% CI 0.28–1.69). Grade ≥ 2 mucositis was observed in 66.7% versus 59.3% of patients (p = 0.41; RR 1.12, 95% CI 0.85–1.48). There was also no difference in the pain scores (p > 0.3). Opioid use was comparable (56.7% vs 61.0%, p = 0.63). Tube feeding was significantly lower in the celecoxib arm (16.7% vs 33.9%, p = 0.03), suggesting better preservation of swallowing function. Both interventions were well tolerated, with mild gastrointestinal side effects. No treatment interruptions or serious adverse events were attributed to the study drugs
Conclusion: Oral celecoxib did not reduce the incidence or severity of acute RIOM compared with glutamine, but may reduce tube-feeding requirements. References: Huang C-J, et al.
