The Blue Pippin is an electrophoresis platform able to separate and collect high molecular weight DNA fragments. Blue Pippin automatically collects target sized DNA in buffer using pre-cast gel cassettes. A maximum of 4 samples per gel cassette can be run, without any cross contamination. Available cassettes allow DNA size selections between 4 kb and 50 kb. This course gives an overview of the Blue Pippin platform specifications based on research requirements, enabling users to successfully use it independently. Sample preparation, software setup, and sample loading will be covered. PARTICIPANT PROFILE This course is intended for students and research professionals from KAUST, and their collaborators from academia and industry, who are involved in genom- ics research and sequencing high molecular weight targeted fragments of DNA. PREREQUISITES • Active Badger account • Laboratory Safety Training • Hazardous Waste Training • Emergency Incident Preparedness Training • User-provided DNA samples with a minimum DNA concentration of 500ng in 30 µL total • Proof of fragment size (gel or Fragment Analyzer), otherwise the sample may be lost MAXIMUM PARTICIPANTS 3 DURATION 2 hours (not including run time) FREQUENCY Twice monthly

The Fragment Analyzer is a parallel capillary electrophoresis system automati- cally performing nucleic acid fragments analysis. Users have the ability to qualify and quantify a wide range of DNA samples in a very short time, calculating frag- ment size and concentration. This course gives users the necessary understanding to independently run the Fragment Analyzer platform. Gel and Sample Preparation, machine set up, and result analysis will be covered.
