Introduction to MALDI-MS Analysis

Monolith NT.115Pico quantifies biomolecular interactions using a Biophysica technology called MicroScale Thermophoresis (MST), which can measure dissociation constants of as low as sub-nanomolar range. This training gives an introduction to interaction analysis with Monolith NT.115Pico. Participants will be given an overview of the system, its operation, experimental parameters settings, sample preparation and run, and data analysis. PARTICIPANT PROFILE This course is intended for students and research professionals from KAUST, and their collaborators from academia and industry, who want to conduct interac- tion analysis with the Monolith NT.115Pico instrument. PREREQUISITES • Active Badger account

Matrix-Assisted Laser Desorption/Ionization (MALDI) is a method of ionization that uses a laser to ionize biomolecules embedded in a solid matrix for subse- quent Mass Spectrometric (MS) analysis. It can be used for the ionization of large molecules with minimal fragmentation. In this training, the users will be introduced to the three-step process of MALDI-MS. First, they will learn how to prepare peptide samples by mixing them with a suitable matrix and applying them to a metal plate or target. The users will then be introduced to the use of a pulsed laser to ionize the sample and record a mass spectrum on a Time-of- flight Mass Spectrometer (TOFMS). Finally, the training will cover basic data search features. PARTICIPANT PROFILE This course is intended for students and research professionals from KAUST, and their collaborators from academia and industry, who want to use MALDI-MS.
